To Study the Culture Media Preferences and Growth Conditions also Attempt for the Quantification of Phytoconstituents Present in Hygrophila Auriculata

ABSTRACT
During “To study the culture media and growth conditions and attempt for the quantification of phytoconstituents present in Hygrophila auriculata” we have collected germplasm from different sites of vindhyan region and of different parts and sections were cut and removed from the plant and fixed in formalin acetic acid solution (formalin: acetic acid: 70% ethyl alcohol in the ratio of 0.5:0.5:9). These germpalsm were taken and proceeds for their tissue culture. For shoot organogenesis, young leaves were used as explants and cultured on Murashige and Skoog (Murashige and Skoog 1962 basal medium supplemented with BA, NAA, IAA, or 2iP at different concentrations. The preliminary HPLC studies revealed that the solvent system n- hexane: ethyl acetate (7:3) was ideal and gave well resolved sample peaks. The spots of the chromatogram were visualized at 366 nm with a 500 k filter at Rf values of 0.07, 0.36, 0.50, 0.57, 0.73, 0.84, 0.94 and 0.97. The densitometric scanning at 366 nm gives five major spots with an area of 16.87, 20.46, 12.26, 22.33 and 31.40% at Rf value of, 0.57, 0.73, 0.84, 0.94 and 0.97 respectively.
